IMPACT OF ENTOMOPATHOGENIC VIRUSES AND BACTERIA ON TARNISHED PLANT BUG, LYGUS LINEOLARIS POPULATIONS

Incidence of Wolbachia in natural populations of L. lineolaris in the Mississippi Delta is being investigated by polymerase chain reaction (PCR) amplification of conserved Wolbachia surface protein (WSP). Genomic DNA and RNA extracted from Lygus lineolaris collected from nine locations in the Mississippi Delta in 2007 and 2009 and from six locations in Pennsylvania in 2007 (45-48 insects from each location) were screened for the presence of Wolbachia. Lygus populations from Vicksburg, Stoneville, and Rolling Fork collected in 2007 had 2-8% infection rates while all other samples collected in 2007 were negative for Wolbachia. Of the 2009 samples screened, 4-6% of insects from Stoneville, Tunica, and Clarksdale insects were infected with Wolbachia. By PCR amplifying, cloning, and sequencing five standard genes used for multi locus genotyping of Wolbachia, we have confirmed the identity of two strains of Wolbachia endosymbionts in Lygus lineolaris from the Delta. Detailed genotyping of insects suspected of harboring a third Wolbachia strain is currently underway. Two Lygus colonies, each infected with a different strain of Wolbachia were established by collecting eggs from a single infected female and inbreeding the progeny. These colonies are being used for reciprocal crosses to study the effects of Wolbachia infections on the reproductive compatibility between infected and uninfected insects as well as insects infected with different strains of Wolbachia.
